Project Description:
Although major advances have been made in knowledge of plant genomes and the recombination processes regulating their structure and integrity, understanding of mechanisms of plant genome dynamics is limited by our relatively poor understanding of plant recombination and the absence of a methodology for the targeted, in situ modification of plant genes and chromosomes. In plants the somatic homologous recombination machinery is overwhelmed by another recombination process (NHEJ), which causes incoming DNA to insert at a random position in the genome, often accompanied by unwanted rearrangements. Solving this via understanding the mechanisms involved and their control is the subject of this project. We have 3 major objectives: (1) develop the means controlled in situ plant gene modification (gene targeting) for phenotyping in functional genome analysis; (2) using mutant plants, improve understanding of and manipulate recombination in plants; (3) test the application of these results to a crop plant. The groups will contribute considerable novel expertise and biological material to the project: plant hyperecombination mutants, cloned genes and proven assays.
